Cleaning water with bacteria

There seems to be much interest in bacteria as collaborators as opposed to the old ‘enemy that must be destoyed’ concept. The latest collaborative effort was announced in a January 19,2019 news item on Nanowerk,

More than one in 10 people in the world lack basic drinking water access, and by 2025, half of the world’s population will be living in water-stressed areas, which is why access to clean water is one of the National Academy of Engineering’s Grand Challenges. Engineers at Washington University in St. Louis [WUSTL] have designed a novel membrane technology that purifies water while preventing biofouling, or buildup of bacteria and other harmful microorganisms that reduce the flow of water.

And they used bacteria to build such filtering membranes.

A January 17, 2019 WUSTL news release by Beth Miller, which originated the news item, provides more detail,

Srikanth Singamaneni, professor of mechanical engineering & materials science, and Young-Shin Jun, professor of energy, environmental & chemical engineering, and their teams blended their expertise to develop an ultrafiltration membrane using graphene oxide and bacterial nanocellulose that they found to be highly efficient, long-lasting and environmentally friendly. If their technique were to be scaled up to a large size, it could benefit many developing countries where clean water is scarce.


Biofouling accounts for nearly half of all membrane fouling and is highly challenging to eradicate completely. Singamaneni and Jun have been tackling this challenge together for nearly five years. They previously developed other membranes using gold nanostars, but wanted to design one that used less expensive materials.

Their new membrane begins with feeding Gluconacetobacter hansenii bacteria a sugary substance so that they form cellulose nanofibers when in water. The team then incorporated graphene oxide (GO) flakes into the bacterial nanocellulose while it was growing, essentially trapping GO in the membrane to make it stable and durable.

After GO is incorporated, the membrane is treated with base solution to kill Gluconacetobacter. During this process, the oxygen groups of GO are eliminated, making it reduced GO.  When the team shone sunlight onto the membrane, the reduced GO flakes immediately generated heat, which is dissipated into the surrounding water and bacteria nanocellulose.

Ironically, the membrane created from bacteria also can kill bacteria.
“If you want to purify water with microorganisms in it, the reduced graphene oxide in the membrane can absorb the sunlight, heat the membrane and kill the bacteria,” Singamaneni said.

Singamaneni and Jun and their team exposed the membrane to E. coli bacteria, then shone light on the membrane’s surface. After being irradiated with light for just 3 minutes, the E. coli bacteria died. The team determined that the membrane quickly heated to above the 70 degrees Celsius required to deteriorate the cell walls of E. coli bacteria.

While the bacteria are killed, the researchers had a pristine membrane with a high quality of nanocellulose fibers that was able to filter water twice as fast as commercially available ultrafiltration membranes under a high operating pressure.

When they did the same experiment on a membrane made from bacterial nanocellulose without the reduced GO, the E. coli bacteria stayed alive.

“This is like 3-D printing with microorganisms,” Jun said. “We can add whatever we like to the bacteria nanocellulose during its growth. We looked at it under different pH conditions similar to what we encounter in the environment, and these membranes are much more stable compared to membranes prepared by vacuum filtration or spin-coating of graphene oxide.”

While Singamaneni and Jun acknowledge that implementing this process in conventional reverse osmosis systems is taxing, they propose a spiral-wound module system, similar to a roll of towels. It could be equipped with LEDs or a type of nanogenerator that harnesses mechanical energy from the fluid flow to produce light and heat, which would reduce the overall cost.

Trounce biofouling with nanowrinkles

This is an example of what the researchers mean by ‘nanowrinkles’,

As for ‘biofouling’, here’s my rough and ready description for anyone who might find it helpful, if you’ve been to the beach and slipped on some rocks, that slip was probably due to a biofilm and biofilm contributes to ‘biofouling’.

Australian scientists have announced research on a new technique for the eradication of biofouling according to a January 17, 2018 University of Sydney press release (also on EurekAlert but dated Jan. 16, 2018),

Sydney scientists have developed nanowrinkled coatings that could avoid the build-up of damaging biological material and save some of the $320 million annually spent by the Australian shipping industry because of biofouling.

A team of chemistry researchers from the University of Sydney Nano Institute has developed nanostructured surface coatings that have anti-fouling properties without using any toxic components.

Biofouling – the build-up of damaging biological material – is a huge economic issue, costing the aquaculture and shipping industries billions of dollars a year in maintenance and extra fuel usage. It is estimated that the increased drag on ship hulls due to biofouling costs the shipping industry in Australia $320 million a year a b.

Since the banning of the toxic anti-fouling agent tributyltin, the need for new non-toxic methods to stop marine biofouling has been pressing.

Leader of the research team, Associate Professor Chiara Neto, said: “We are keen to understand how these surfaces work and also push the boundaries of their application, especially for energy efficiency. Slippery coatings are expected to be drag-reducing, which means that objects, such as ships, could move through water with much less energy required.”

The new materials were tested tied to shark netting in Sydney’s Watson Bay, showing that the nanomaterials were efficient at resisting biofouling in a marine environment.

The research has been published in ACS Applied Materials & Interfaces.

The new coating uses ‘nanowrinkles’ inspired by the carnivorous Nepenthes pitcher plant. The plant traps a layer of water on the tiny structures around the rim of its opening. This creates a slippery layer causing insects to aquaplane on the surface, before they slip into the pitcher where they are digested.

Nanostructures utilise materials engineered at the scale of billionths of a metre – 100,000 times smaller than the width of a human hair. Associate Professor Neto’s group at Sydney Nano is developing nanoscale materials for future development in industry.

Biofouling can occur on any surface that is wet for a long period of time, for example aquaculture nets, marine sensors and cameras, and ship hulls. The slippery surface developed by the Neto group stops the initial adhesion of bacteria, inhibiting the formation of a biofilm from which larger marine fouling organisms can grow.

The interdisciplinary University of Sydney team included biofouling expert Professor Truis Smith-Palmer of St Francis Xavier University in Nova Scotia, Canada, who was on sabbatical visit to the Neto group for a year, partially funded by the Faculty of Science scheme for visiting women.

In the lab, the slippery surfaces resisted almost all fouling from a common species of marine bacteria, while control Teflon samples without the lubricating layer were completely fouled. Not satisfied with testing the surfaces under highly controlled lab conditions with only one type of bacteria the team also tested the surfaces in the ocean, with the help of marine biologist Professor Ross Coleman.

Test surfaces were attached to swimming nets at Watsons Bay baths in Sydney Harbour for a period of seven weeks. In the much harsher marine environment, the slippery surfaces were still very efficient at resisting fouling.

The antifouling coatings are mouldable and transparent, making their application ideal for underwater cameras and sensors.

Slip sliding away—making surfaces bacteria can’t grasp onto

Here’s another biomimicry story with a connection to Harvard University. From a Nov. 1, 2016 Beth Israel Deaconess Medical Center (Harvard Medical School Teaching Hospital) news release (also on EurekAlert),

Implanted medical devices like catheters, surgical mesh and dialysis systems are ideal surfaces on which bacteria can colonize and form hard-to-kill sheets called biofilms. Known as biofouling, this contamination of devices is responsible for more than half of the 1.7 million hospital-acquired infections in the United States each year.

In a report published in Biomaterials today, a team of scientists at Beth Israel Deaconess Medical Center (BIDMC), the Wyss Institute for Biologically Inspired Engineering and the John A. Paulson School of Engineering and Applied Sciences (SEAS) at Harvard University has demonstrated that an innovative, ultra-low adhesive coating prevented bacteria from attaching to surfaces treated with it, reducing bacterial adhesion by more than 98 percent in laboratory tests.

“Device related infections remain a significant problem in medicine, burdening society with millions of dollars in health care costs,” said Elliot Chaikof, MD, PhD, chair of the Roberta and Stephen R. Weiner Department of Surgery and Surgeon-in-Chief at BIDMC and an associate faculty member at the Wyss Institute. “Antibiotics alone will not solve this problem. We need to use new approaches to minimize the risk of infection, and this strategy is a very important step in that direction.”

The self-healing slippery surface coatings – known as ‘slippery liquid-infused porous surfaces’ (SLIPS) – were developed by Joanna Aizenberg, PhD, a Wyss Institute core faculty member, Professor of Chemistry and Chemical Biology and the Amy Smith Berylson Professor of Materials Science at SEAS at Harvard University. Inspired by the carnivorous Nepenthes pitcher plant that uses the slippery surface of its leaves to trap insects, Aizenberg engineered surface coatings that work to repel a variety of substances across a broad range of temperature, pressure and other environmental conditions. They are stable when exposed to UV light, and are low-cost and simple to manufacture. The current study is the first to demonstrate that SLIPS not only limit the ability of bacteria to adhere to surfaces, but also impede infection in an animal model.

SLIPS has been mentioned here before, most recently in a March 2, 2016 posting and before that in an Oct. 14, 2014 posting which appears to be precursor work for this latest research.

Getting back to the Nov. 1, 2016 news release, here’s more about plans for SLIPS and about recent trials,

“We are developing SLIPS recipes for a variety of medical applications by working with different medical-grade materials, ensuring the stability of the coating, and carefully pairing the non-fouling properties of the SLIPS materials to specific contaminates, environments and performance requirements,” said Aizenberg. “Here we have extended our repertoire and applied the SLIPS concept very convincingly to medical-grade lubricants, demonstrating its enormous potential in implanted devices prone to bacterial fouling and infection.”

In a series of trials, the researchers tested three SLIPS lubricants for their anti-adhesive qualities. First, they incubated disks of SLIPS-coated medical material ePTFE – a microporous form of Teflon – in a broth of Staphylococcus aureus (S. aureus), a generally harmless bacterium found in the nose and on skin, but one of the most common causes of hospital-acquired infections. After 48 hours, the three variations of SLIPS-treated disks demonstrated 98.3, 99.1 and 99.7 percent reductions in bacterial adhesion.

To test the material’s stability, the scientists performed the same experiment after soaking the SLIPS-coated samples for up to 21 days in a solution meant to simulate conditions inside a living mammal. After exposing these disks to S. aureus for 48 hours, the researchers found similar, nearly 100 percent reductions in bacterial adhesion.

Widely used clinically, medical mesh is particularly susceptible to bacterial infection. In another set of experiments to test the material’s biocompatibility, Chaikof and colleagues implanted small squares of SLIPS-treated mesh into murine models, injecting the site with S. aureus 24 hours later. Three days later, when the researchers removed the implanted mesh, they found little to no infection, compared with an infection rate of more than 90 percent among controls.

“Today, patients who receive implants often require antibiotics to keep the risk of bacterial infection at bay,” the authors wrote. “SLIPS coatings one day could obviate the widespread use of antibiotics and minimize the development of antibiotic resistant micro-organisms.”

“SLIPs have many promising medical applications that are in a very early stage of evaluation,” said Chaikof. “Clearly, there’s more work to be done before its introduction into the clinic, but this is one of a few studies that reinforces the exciting opportunities presented by this strategy to improve device performance and clinical outcomes.”

No more boat scraping with new coating from Duke University

There’s a lot of interest in finding ways to discourage bacteria from growing on various surfaces, for example, Sharklet, which is based on nanostructures on sharkskin, is a product being developed for hospitals (my Feb. 10, 2011 posting) and there are polymers that ‘uninvite’ bacteria at the University of Nottingham (my Aug. 13, 2012 posting).

A Jan. 31, 2013 news item on Nanowerk highlights the latest work being done at Duke University,

Duke University engineers have developed a material that can be applied like paint to the hull of a ship and will literally be able to dislodge bacteria, keeping it from accumulating on the ship’s surface. This buildup on ships increases drag and reduces the energy efficiency of the vessel, as well as blocking or clogging undersea sensors.

Dynamic change of surface area and topology of elastomers is used as a general, environmentally friendly approach for effectively detaching micro- and macro-fouling organisms adhered on the elastomer surfaces. Deformation of elastomer surfaces under electrical or pneumatic actuation can debond various biofilms and barnacles. The bio-inspired dynamic surfaces can be fabricated over large areas through simple and practical processes. This new mechanism is complementary with existing materials and methods for biofouling control.

Duke University’s Jan. 31, 2013 news release by Richard Merritt, which originated the news item, provides more detail from the researchers,

“We have developed a material that ‘wrinkles,’ or changes it surface in response to a stimulus, such as stretching or pressure or electricity,” said Duke engineer Xuanhe Zhao, assistant professor in Duke’s Pratt School of Engineering. “This deformation can effectively detach biofilms and other organisms that have accumulated on the surface.”

Zhao has already demonstrated the ability of electric current to deform, or change, the surface of polymers.

The researchers tested their approach in the laboratory with simulated seawater, as well as on barnacles. These experiments were conducted in collaboration with Daniel Rittsch of the Duke University Marine Lab in Beaufort, N.C.

Keeping bacteria from attaching to ship hulls or other submerged objects can prevent a larger cascade of events that can reduce performance or efficiency. Once they have taken up residence on a surface, bacteria often attract larger organisms, such as seaweed and larva of other marine organisms, such as worms, bivalves, barnacles or mussels.
