Mini T-shirt demonstrates photosynthetic living materials

I’m not sure how I feel about a t-shirt, regardless of size, made of living biological material but these researchers seem uniformly enthusiastic. From a May 3, 2021 news item on phys.org (Note: A link has been removed),

Living materials, which are made by housing biological cells within a non-living matrix, have gained popularity in recent years as scientists recognize that often the most robust materials are those that mimic nature.

For the first time, an international team of researchers from the University of Rochester [located in New York state, US] and Delft University of Technology in the Netherlands used 3D printers and a novel bioprinting technique to print algae into living, photosynthetic materials that are tough and resilient. The material has a variety of applications in the energy, medical, and fashion sectors. The research is published in the journal Advanced Functional Materials.

An April 30, 2021 University of Rochester new release (also on EurekAlert but published May 3, 2021) by Lindsey Valich, which originated the news item, delves further into the topic of living materials,

“Three-dimensional printing is a powerful technology for fabrication of living functional materials that have a huge potential in a wide range of environmental and human-based applications.” says Srikkanth Balasubramanian, a postdoctoral research associate at Delft and the first author of the paper. “We provide the first example of an engineered photosynthetic material that is physically robust enough to be deployed in real-life applications.”

HOW TO BUILD NEW MATERIALS: LIVING AND NONLIVING COMPONENTS

To create the photosynthetic materials, the researchers began with a non-living bacterial cellulose–an organic compound that is produced and excreted by bacteria. Bacterial cellulose has many unique mechanical properties, including its flexibility, toughness, strength, and ability to retain its shape, even when twisted, crushed, or otherwise physically distorted.

The bacterial cellulose is like the paper in a printer, while living microalgae acts as the ink. The researchers used a 3D printer to deposit living algae onto the bacterial cellulose.

The combination of living (microalgae) and nonliving (bacterial cellulose) components resulted in a unique material that has the photosynthetic quality of the algae and the robustness of the bacterial cellulose; the material is tough and resilient while also eco-friendly, biodegradable, and simple and scalable to produce. The plant-like nature of the material means it can use photosynthesis to “feed” itself over periods of many weeks, and it is also able to be regenerated–a small sample of the material can be grown on-site to make more materials.

ARTIFICIAL LEAVES, PHOTOSYNTHETIC SKINS, AND BIO-GARMENTS

The unique characteristics of the material make it an ideal candidate for a variety of applications, including new products such as artificial leaves, photosynthetic skins, or photosynthetic bio-garments.

Artificial leaves are materials that mimic actual leaves in that they use sunlight to convert water and carbon dioxide–a major driver of climate change–into oxygen and energy, much like leaves during photosynthesis. The leaves store energy in chemical form as sugars, which can then be converted into fuels. Artificial leaves therefore offer a way to produce sustainable energy in places where plants don’t grow well, including outer space colonies. The artificial leaves produced by the researchers at Delft and Rochester are additionally made from eco-friendly materials, in contrast to most artificial leaf technologies currently in production, which are produced using toxic chemical methods.

“For artificial leaves, our materials are like taking the ‘best parts’ of plants–the leaves–which can create sustainable energy, without needing to use resources to produce parts of plants–the stems and the roots–that need resources but don’t produce energy,” says Anne S. Meyer, an associate professor of biology at Rochester. “We are making a material that is only focused on the sustainable production of energy.”

Another application of the material would be photosynthetic skins, which could be used for skin grafts, Meyer says. “The oxygen generated would help to kick-start healing of the damaged area, or it might be able to carry out light-activated wound healing.”

Besides offering sustainable energy and medical treatments, the materials could also change the fashion sector. Bio-garments made from algae would address some of the negative environmental effects of the current textile industry in that they would be high-quality fabrics that would be sustainability produced and completely biodegradable. They would also work to purify the air by removing carbon dioxide through photosynthesis and would not need to be washed as often as conventional garments, reducing water usage.

“Our living materials are promising because they can survive for several days with no water or nutrients access, and the material itself can be used as a seed to grow new living materials,” says Marie-Eve Aubin-Tam, an associate professor of bionanoscience at Delft. “This opens the door to applications in remote areas, even in space, where the material can be seeded on site.”

Use kombucha to produce bacterial cellulose

The combination of the US Army, bacterial cellulose, and kombucha seems a little unusual. However, this January 26, 2021 U.S. Army Research Laboratory news release (also on EurekAlert) provides some clues as to how this combination makes sense,

Kombucha tea, a trendy fermented beverage, inspired researchers to develop a new way to generate tough, functional materials using a mixture of bacteria and yeast similar to the kombucha mother used to ferment tea.

With Army funding, using this mixture, also called a SCOBY, or symbiotic culture of bacteria and yeast, engineers at MIT [Massachusetts Institute of Technology] and Imperial College London produced cellulose embedded with enzymes that can perform a variety of functions, such as sensing environmental pollutants and self-healing materials.

The team also showed that they could incorporate yeast directly into the cellulose, creating living materials that could be used to purify water for Soldiers in the field or make smart packaging materials that can detect damage.

“This work provides insights into how synthetic biology approaches can harness the design of biotic-abiotic interfaces with biological organization over multiple length scales,” said Dr. Dawanne Poree, program manager, Army Research Office, an element of the U.S. Army Combat Capabilities Development Command, now known as DEVCOM, Army Research Laboratory. “This is important to the Army as this can lead to new materials with potential applications in microbial fuel cells, sense and respond systems, and self-reporting and self-repairing materials.”

The research, published in Nature Materials was funded by ARO [Army Research Office] and the Army’s Institute for Soldier Nanotechnologies [ISN] at the Massachusetts Institute of Technology. The U.S. Army established the ISN in 2002 as an interdisciplinary research center devoted to dramatically improving the protection, survivability, and mission capabilities of the Soldier and Soldier-supporting platforms and systems.

“We foresee a future where diverse materials could be grown at home or in local production facilities, using biology rather than resource-intensive centralized manufacturing,” said Timothy Lu, an MIT associate professor of electrical engineering and computer science and of biological engineering.

Researchers produced cellulose embedded with enzymes, creating living materials that could be used to purify water for Soldiers in the field or make smart packaging materials that can detect damage. These fermentation factories, which usually contain one species of bacteria and one or more yeast species, produce ethanol, cellulose, and acetic acid that gives kombucha tea its distinctive flavor.

Most of the wild yeast strains used for fermentation are difficult to genetically modify, so the researchers replaced them with a strain of laboratory yeast called Saccharomyces cerevisiae. They combined the yeast with a type of bacteria called Komagataeibacter rhaeticus that their collaborators at Imperial College London had previously isolated from a kombucha mother. This species can produce large quantities of cellulose.

Because the researchers used a laboratory strain of yeast, they could engineer the cells to do any of the things that lab yeast can do, such as producing enzymes that glow in the dark, or sensing pollutants or pathogens in the environment. The yeast can also be programmed so that they can break down pollutants/pathogens after detecting them, which is highly relevant to Army for chem/bio defense applications.

“Our community believes that living materials could provide the most effective sensing of chem/bio warfare agents, especially those of unknown genetics and chemistry,” said Dr. Jim Burgess ISN program manager for ARO.

The bacteria in the culture produced large-scale quantities of tough cellulose that served as a scaffold. The researchers designed their system so that they can control whether the yeast themselves, or just the enzymes that they produce, are incorporated into the cellulose structure. It takes only a few days to grow the material, and if left long enough, it can thicken to occupy a space as large as a bathtub.

“We think this is a good system that is very cheap and very easy to make in very large quantities,” said MIT graduate student and the paper’s lead author, Tzu-Chieh Tang. To demonstrate the potential of their microbe culture, which they call Syn-SCOBY, the researchers created a material incorporating yeast that senses estradiol, which is sometimes found as an environmental pollutant. In another version, they used a strain of yeast that produces a glowing protein called luciferase when exposed to blue light. These yeasts could be swapped out for other strains that detect other pollutants, metals, or pathogens.

The researchers are now looking into using the Syn-SCOBY system for biomedical or food applications. For example, engineering the yeast cells to produce antimicrobials or proteins that could benefit human health.

Semi-living gloves as sensors

Researchers at the Massachusetts Institute of Technology (MIT) are calling it a new ‘living material’ according to a Feb. 16, 2017 news item on Nanowerk,

Engineers and biologists at MIT have teamed up to design a new “living material” — a tough, stretchy, biocompatible sheet of hydrogel injected with live cells that are genetically programmed to light up in the presence of certain chemicals.

In a paper published this week in the Proceedings of the National Academy of Sciences, the researchers demonstrate the new material’s potential for sensing chemicals, both in the environment and in the human body.

The team fabricated various wearable sensors from the cell-infused hydrogel, including a rubber glove with fingertips that glow after touching a chemically contaminated surface, and bandages that light up when pressed against chemicals on a person’s skin.

Xuanhe Zhao, the Robert N. Noyce Career Development associate professor of mechanical engineering at MIT, says the group’s living material design may be adapted to sense other chemicals and contaminants, for uses ranging from crime scene investigation and forensic science, to pollution monitoring and medical diagnostics.

“With this design, people can put different types of bacteria in these devices to indicate toxins in the environment, or disease on the skin,” says Timothy Lu, associate professor of biological engineering and of electrical engineering and computer science. “We’re demonstrating the potential for living materials and devices.”

The paper’s co-authors are graduate students Xinyue Liu, Tzu-Chieh Tang, Eleonore Tham, Hyunwoo Yuk, and Shaoting Lin.

Infusing life in materials

Lu and his colleagues in MIT’s Synthetic Biology Group specialize in creating biological circuits, genetically reprogramming the biological parts in living cells such as E. coli to work together in sequence, much like logic steps in an electrical circuit. In this way, scientists can reengineer living cells to carry out specific functions, including the ability to sense and signal the presence of viruses and toxins.

However, many of these newly programmed cells have only been demonstrated in situ, within Petri dishes, where scientists can carefully control the nutrient levels necessary to keep the cells alive and active — an environment that has proven extremely difficult to replicate in synthetic materials.

“The challenge to making living materials is how to maintain those living cells, to make them viable and functional in the device,” Lu says. “They require humidity, nutrients, and some require oxygen. The second challenge is how to prevent them from escaping from the material.”

To get around these roadblocks, others have used freeze-dried chemical extracts from genetically engineered cells, incorporating them into paper to create low-cost, virus-detecting diagnostic strips. But extracts, Lu says, are not the same as living cells, which can maintain their functionality over a longer period of time and may have higher sensitivity for detecting pathogens.

Other groups have seeded heart muscle cells onto thin rubber films to make soft, “living” actuators, or robots. When bent repeatedly, however, these films can crack, allowing the live cells to leak out.

A lively host

Zhao’s group in MIT’s Soft Active Materials Laboratory has developed a material that may be ideal for hosting living cells. For the past few years, his team has come up with various formulations of hydrogel — a tough, highly stretchable, biocompatible material made from a mix of polymer and water. Their latest designs have contained up to 95 percent water, providing an environment which Zhao and Lu recognized might be suitable for sustaining living cells. The material also resists cracking even when repeatedly stretched and pulled — a property that could help contain cells within the material.

The two groups teamed up to integrate Lu’s genetically programmed bacterial cells into Zhao’s sheets of hydrogel material. They first fabricated layers of hydrogel and patterned narrow channels within the layers using 3-D printing and micromolding techniques. They fused the hydrogel to a layer of elastomer, or rubber, that is porous enough to let in oxygen. They then injected E. coli cells into the hydrogel’s channels. The cells were programmed to fluoresce, or light up, when in contact with certain chemicals that pass through the hydrogel, in this case a natural compound known as DAPG (2,4-diacetylphloroglucinol).

The researchers then soaked the hydrogel/elastomer material in a bath of nutrients which infused throughout the hydrogel and helped to keep the bacterial cells alive and active for several days.

To demonstrate the material’s potential uses, the researchers first fabricated a sheet of the material with four separate, narrow channels, each containing a type of bacteria engineered to glow green in response to a different chemical compound. They found each channel reliably lit up when exposed to its respective chemical.

Next, the team fashioned the material into a bandage, or “living patch,” patterned with channels containing bacteria sensitive to rhamnose, a naturally occurring sugar. The researchers swabbed a volunteer’s wrist with a cotton ball soaked in rhamnose, then applied the hydrogel patch, which instantly lit up in response to the chemical.

Finally, the researchers fabricated a hydrogel/elastomer glove whose fingertips contained swirl-like channels, each of which they filled with different chemical-sensing bacterial cells. Each fingertip glowed in response to picking up a cotton ball soaked with a respective compound.

The group has also developed a theoretical model to help guide others in designing similar living materials and devices.

“The model helps us to design living devices more efficiently,” Zhao says. “It tells you things like the thickness of the hydrogel layer you should use, the distance between channels, how to pattern the channels, and how much bacteria to use.”

Ultimately, Zhao envisions products made from living materials, such as gloves and rubber soles lined with chemical-sensing hydrogel, or bandages, patches, and even clothing that may detect signs of infection or disease.

Do you have a proposal for living building materials?

DARPA (US Defense Advanced Research Projects Agency) has launched a program called Engineered Living Materials (ELM) and issued an invitation. From an Aug. 9, 2016 news item on Nanowerk,

The structural materials that are currently used to construct homes, buildings, and infrastructure are expensive to produce and transport, wear out due to age and damage, and have limited ability to respond to changes in their immediate surroundings. Living biological materials—bone, skin, bark, and coral, for example—have attributes that provide advantages over the non-living materials people build with, in that they can be grown where needed, self-repair when damaged, and respond to changes in their surroundings. The inclusion of living materials in human-built environments could offer significant benefits; however, today scientists and engineers are unable to easily control the size and shape of living materials in ways that would make them useful for construction.

DARPA is launching the Engineered Living Materials (ELM) program with a goal of creating a new class of materials that combines the structural properties of traditional building materials with attributes of living systems. Living materials represent a new opportunity to leverage engineered biology to solve existing problems associated with the construction and maintenance of built environments, and to create new capabilities to craft smart infrastructure that dynamically responds to its surroundings.

“The vision of the ELM program is to grow materials on demand where they are needed,” said ELM program manager Justin Gallivan. “Imagine that instead of shipping finished materials, we can ship precursors and rapidly grow them on site using local resources. And, since the materials will be alive, they will be able to respond to changes in their environment and heal themselves in response to damage.”

Grown materials are not entirely new, but their current manifestations differ substantially from the materials Gallivan envisions. For instance, biologically sourced structural materials can already be grown into specified sizes and shapes from inexpensive feedstocks; packing materials derived from fungal mycelium and building blocks made from bacteria and sand are two modern examples. And, of course, wood has been used for ages. However, these products are rendered inert during the manufacturing process, so they exhibit few of their components’ original biological advantages. Scientists are making progress with three-dimensional printing of living tissues and organs, using scaffolding materials that sustain the long-term viability of the living cells. These cells are derived from existing natural tissues, however, and are not engineered to perform synthetic functions. And current cell-printing methods are too expensive to produce building materials at necessary scales.

ELM looks to merge the best features of these existing technologies and build on them to create hybrid materials composed of non-living scaffolds that give structure to and support the long-term viability of engineered living cells. DARPA intends to develop platform technologies that are scalable and generalizable to facilitate a quick transition from laboratory to commercial applications.

The long-term objective of the ELM program is to develop an ability to engineer structural properties directly into the genomes of biological systems so that neither scaffolds nor external development cues are needed for an organism to realize the desired shape and properties. Achieving this goal will require significant breakthroughs in scientists’ understanding of developmental pathways and how those pathways direct the three-dimensional development of multicellular systems.

Work on ELM will be fundamental research carried out in controlled laboratory settings. DARPA does not anticipate environmental release during the program.

3) To encourage and promote teaming arrangements among organizations that have the relevant expertise, research facilities and capabilities for executing research and development responsive to the ELM program goals.

The Proposers Day will include overview presentations and optional sidebar meetings where potential proposers can discuss ideas for proposal submissions with the Government team.

The goal of the DARPA ELM program is to explore and develop living materials that combine the structural properties of traditional building materials with attributes of living systems, including the ability to rapidly grow, to self-repair, and to adapt to the environment. Living materials represent a new opportunity to leverage engineered biology to solve existing problems associated with the construction and maintenance of our built environments, as well as to create new capabilities to craft smart infrastructure that dynamically responds to our surroundings. The specific program objectives are to develop design tools and methods that enable the engineering of structural features into cellular systems that function as living materials, thereby opening up a new design space for building technology. These new methods will be validated by the production of living materials that can reproduce, self-organize and self-heal.
