Keeping up with science is impossible: ruminations on a nanotechnology talk

I think it’s time to give this suggestion again. Always hold a little doubt about the science information you read and hear. Everybody makes mistakes.

Here’s an example of what can happen. George Tulevski who gave a talk about nanotechnology in Nov. 2016 for TED@IBM is an accomplished scientist who appears to have made an error during his TED talk. From Tulevski’s The Next Step in Nanotechnology talk transcript page,

When I was a graduate student, it was one of the most exciting times to be working in nanotechnology. There were scientific breakthroughs happening all the time. The conferences were buzzing, there was tons of money pouring in from funding agencies. And the reason is when objects get really small, they’re governed by a different set of physics that govern ordinary objects, like the ones we interact with. We call this physics quantum mechanics. [emphases mine] And what it tells you is that you can precisely tune their behavior just by making seemingly small changes to them, like adding or removing a handful of atoms, or twisting the material. It’s like this ultimate toolkit. You really felt empowered; you felt like you could make anything.

In September 2016, scientists at Cambridge University (UK) announced they had concrete proof that the physics governing materials at the nanoscale is unique, i.e., it does not follow the rules of either classical or quantum physics. From my Oct. 27, 2016 posting,

In the middle, on the order of around 10–100,000 molecules, something different is going on. Because it’s such a tiny scale, the particles have a really big surface-area-to-volume ratio. This means the energetics of what goes on at the surface become very important, much as they do on the atomic scale, where quantum mechanics is often applied.

Classical thermodynamics breaks down. But because there are so many particles, and there are many interactions between them, the quantum model doesn’t quite work either.

It is very, very easy to miss new developments no matter how tirelessly you scan for information.

Tulevski is a good, interesting, and informed speaker but I do have one other hesitation regarding his talk. He seems to think that over the last 15 years there should have been more practical applications arising from the field of nanotechnology. There are two aspects here. First, he seems to be dating the ‘nanotechnology’ effort from the beginning of the US National Nanotechnology Initiative and there are many scientists who would object to that as the starting point. Second, 15 or even 30 or more years is a brief period of time especially when you are investigating that which hasn’t been investigated before. For example, you might want to check out the book, “Leviathan and the Air-Pump: Hobbes, Boyle, and the Experimental Life” (published 1985) is a book by Steven Shapin and Simon Schaffer (Wikipedia entry for the book). The amount of time (years) spent on how to make just the glue which held the various experimental apparatuses together was a revelation to me. Of  course, it makes perfect sense that if you’re trying something new, you’re going to have figure out everything.

By the way, I include my blog as one of the sources of information that can be faulty despite efforts to make corrections and to keep up with the latest. Even the scientists at Cambridge University can run into some problems as I noted in my Jan. 28, 2016 posting.

The brief description of this odyssey includes US $78 million in financing over 15 years and $50 million in revenues over that period through licensing of its technology and patents. That revenue includes a back-against-the-wall sell-off of a key business unit to Lockheed Martin in 2008.  Another key moment occured back in 2012 when Belgian-based nanoelectronics powerhouse Imec took on the job of further developing Nantero’s carbon-nanotube-based memory back in 2012. Despite the money and support from major electronics players, the big commercial breakout of their NRAM technology seemed ever less likely to happen with the passage of time.

A view to controversies about nanoparticle drug delivery, sticky-flares, and a PNAS surprise

Despite all the excitement and claims for nanoparticles as vehicles for drug delivery to ‘sick’ cells there is at least one substantive problem, the drug-laden nanoparticles don’t actually enter the interior of the cell. They are held in a kind of cellular ‘waiting room’.

Leonid Schneider in a Nov. 20, 2015 posting on his For Better Science blog describes the process in more detail,

A large body of scientific nanotechnology literature is dedicated to the biomedical aspect of nanoparticle delivery into cells and tissues. The functionalization of the nanoparticle surface is designed to insure their specificity at targeting only a certain type of cells, such as cancers cells. Other technological approaches aim at the cargo design, in order to ensure the targeted release of various biologically active agents: small pharmacological substances, peptides or entire enzymes, or nucleotides such as regulatory small RNAs or even genes. There is however a main limitation to this approach: though cells do readily take up nanoparticles through specific membrane-bound receptor interaction (endocytosis) or randomly (pinocytosis), these nanoparticles hardly ever truly reach the inside of the cell, namely its nucleocytoplasmic space. Solid nanoparticles are namely continuously surrounded by the very same membrane barrier they first interacted with when entering the cell. These outer-cell membrane compartments mature into endosomal and then lysosomal vesicles, where their cargo is subjected to low pH and enzymatic digestion. The nanoparticles, though seemingly inside the cell, remain actually outside. …

What follows is a stellar piece featuring counterclaims about and including Schneider’s own journalistic research into scientific claims that the problem of gaining entry to a cell’s true interior has been addressed by technologies developed in two different labs.

Having featured one of the technologies here in a July 24, 2015 posting titled: Sticky-flares nanotechnology to track and observe RNA (ribonucleic acid) regulation and having been contacted a couple of times by one of the scientists, Raphaël Lévy from the University of Liverpool (UK), challenging the claims made (Lévy’s responses can be found in the comments section of the July 2015 posting), I thought a followup of sorts was in order.

Scientific debates (then and now)

Scientific debates and controversies are part and parcel of the scientific process and what most outsiders, such as myself, don’t realize is how fraught it is. For a good example from the past, there’s Leviathan and the Air-Pump: Hobbes, Boyle, and the Experimental Life (from its Wikipedia entry), Note: Links have been removed),

Leviathan and the Air-Pump: Hobbes, Boyle, and the Experimental Life (published 1985) is a book by Steven Shapin and Simon Schaffer. It examines the debate between Robert Boyle and Thomas Hobbes over Boyle’s air-pump experiments in the 1660s.

The style seems more genteel than what a contemporary Canadian or US audience is accustomed to but Hobbes and Boyle (and proponents of both sides) engaged in bruising communication.

There was a lot at stake then and now. It’s not just the power, prestige, and money, as powerfully motivating as they are, it’s the research itself. Scientists work for years to achieve breakthroughs or to add more to our common store of knowledge. It’s painstaking and if you work at something for a long time, you tend to be invested in it. Saying you’ve wasted ten years of your life looking at the problem the wrong way or have misunderstood your data is not easy.

As for the current debate, Schneider’s description gives no indication that there is rancour between any of the parties but it does provide a fascinating view of two scientists challenging one of the US’s nanomedicine rockstars, Chad Mirkin. The following excerpt follows the latest technical breakthroughs to the interior portion of the cell through three phases of the naming conventions (Nano-Flares, also known by its trade name, SmartFlares, which is a precursor technology to Sticky-Flares), Note: Links have been removed,

The next family of allegedly nucleocytoplasmic nanoparticles which Lévy turned his attention to, was that of the so called “spherical nucleic acids”, developed in the lab of Chad Mirkin, multiple professor and director of the International Institute for Nanotechnology at the Northwestern University, USA. These so called “Nano-Flares” are gold nanoparticles, functionalized with fluorophore-coupled oligonucleotides matching the messenger RNA (mRNA) of interest (Prigodich et al., ACS Nano 3:2147-2152, 2009; Seferos et al., J Am. Chem.Soc. 129:15477-15479, 2007). The mRNA detection method is such that the fluorescence is initially quenched by the gold nanoparticle proximity. Yet when the oligonucleotide is displaced by the specific binding of the mRNA molecules present inside the cell, the fluorescence becomes detectable and serves thus as quantitative read-out for the intracellular mRNA abundance. Exactly this is where concerns arise. To find and bind mRNA, spherical nucleic acids must leave the endosomal compartments. Is there any evidence that Nano-Flares ever achieve this and reach intact the nucleocytoplasmatic space, where their target mRNA is?

Lévy’s lab has focused its research on the commercially available analogue of the Nano-Flares, based on the patent to Mirkin and Northwestern University and sold by Merck Millipore under the trade name of SmartFlares. These were described by Mirkin as “a powerful and prolific tool in biology and medical diagnostics, with ∼ 1,600 unique forms commercially available today”. The work, led by Lévy’s postdoctoral scientist David Mason, now available in post-publication process at ScienceOpen and on Figshare, found no experimental evidence for SmartFlares to be ever found outside the endosomal membrane vesicles. On the contrary, the analysis by several complementary approaches, i.e., electron, fluorescence and photothermal microscopy, revealed that the probes are retained exclusively within the endosomal compartments.

In fact, even Merck Millipore was apparently well aware of this problem when the product was developed for the market. As I learned, Merck performed a number of assays to address the specificity issue. Multiple hundred-fold induction of mRNA by biological cell stimulation (confirmed by quantitative RT-PCR) led to no significant changes in the corresponding SmartFlare signal. Similarly, biological gene downregulation or experimental siRNA knock-down had no effect on the corresponding SmartFlare fluorescence. Cell lines confirmed as negative for a certain biomarker proved highly positive in a SmartFlare assay.  Live cell imaging showed the SmartFlare signal to be almost entirely mitochondrial, inconsistent with reported patterns of the respective mRNA distributions.  Elsewhere however, cyanine dye-labelled oligonucleotides were found to unspecifically localise to mitochondria   (Orio et al., J. RNAi Gene Silencing 9:479-485, 2013), which might account to the often observed punctate Smart Flare signal.

More recently, Mirkin lab has developed a novel version of spherical nucleic acids, named Sticky-Flares (Briley et al., PNAS 112:9591-9595, 2015), which has also been patented for commercial use. The claim is that “the Sticky-flare is capable of entering live cells without the need for transfection agents and recognizing target RNA transcripts in a sequence-specific manner”. To confirm this, Lévy used the same approach as for the striped nanoparticles [not excerpted here]: he approached Mirkin by email and in person, requesting the original microscopy data from this publication. As Mirkin appeared reluctant, Lévy invoked the rules for data sharing by the journal PNAS, the funder NSF as well as the Northwestern University. After finally receiving Mirkin’s thin-optical microscopy data by air mail, Lévy and Mason re-analyzed it and determined the absence of any evidence for endosomal escape, while all Sticky-Flare particles appeared to be localized exclusively inside vesicular membrane compartments, i.e., endosomes (Mason & Levy, bioRxiv 2015).

The PNAS surprise

PNAS (Proceedings of the National Academy of Science) published one of Mirkin’s papers on ‘Sticky-flares’ and is where scientists, Raphaël Lévy and David Mason, submitted a letter outlining their concerns with the ‘Sticky-flares’ research. Here’s the response as reproduced in Lévy’s Nov. 16, 2015 posting on his Rapha-Z-Lab blog

Conclusions

My primary interest in this story is in the view it provides of the scientific process and the importance of and difficulty associated with the debates.

I can’t venture an opinion about the research or the counterarguments other than to say that Lévy’s and Mason’s thoughtful challenge bears more examination than PNAS is inclined to accord. If their conclusions or Chad Mirkin’s are wrong, let that be determined in an open process.

Situating Science and the future

The end is in sight (2014) for Canada’s Situating Science; Science in Human Contexts network or rather,  the organization’s funding from the Social Sciences and Humanities Research Council (SSHRC) will be exhausted sometime soon. According to their Fall 2013 newsletter, they are making plans for the future,

I. SUSTAINING THE NETWORK AND ACTIVITIES BEYOND 2014
While this year is the last for the Situating Science SSHRC Strategic Knowledge Cluster, it is an opportunity to celebrate and build upon our successes. As part of our plans, we will follow up on last year’s “think-tank” and management meetings to set out concrete plans for sustaining the network and activities of Cluster scholars beyond its 7 years. A number of Cluster partners and stakeholders will meet during a second “think-tank” to discuss best strategies for moving forward.

The “think-tank” will dovetail nicely with a special symposium in Ottawa on Science and Society Oct. 21-23. For this symposium, the Cluster is partnering with the Institute for Science, Society and Policy to bring together scholars from various disciplines, public servants and policy workers to discuss key issues at the intersection of science and society. [emphasis mine]  The discussions will be compiled in a document to be shared with stakeholders and the wider public.

The team will continue to seek support and partnerships for projects within the scope of its objectives. Among our top priorities are a partnership to explore sciences, technologies and their publics as well as new partnerships to build upon exchanges between scholars and institutions in India, Singapore and Canada.

There’s not much information about the Science & Society symposium (mentioned in the excerpt from the newsletter)  being held Oct. 21-23, 2013 in Ottawa other than this, from the About page (the text seems as if it was lifted out of a grant proposal),

What?

The Mission of the symposium is to create an open forum, in the Nation’s capital, to understand and address the key issues at the interface of science, technology, society and policy. The event will display the importance of connecting disparate themes and will bring together groups not usually in contact to discuss subjects of common interest and brainstorm solutions to common challenges. It will demonstrate that collaboration among academics, students, policy makers, stakeholders and the public at large can lead to new insights, new perspectives, and a deeper understanding of the social implications of science and technology.  It will also make the discussion of science more prominent in the national dialogue.

The symposium will be a major event in Ottawa during National Science and Technology Week. It is a collaboration between the Situating Science Strategic Knowledge Cluster and Institute for Science, Society and Policy (ISSP).

Fostering dialogue between scholars, students, public servants and the general public will not only shed new light on the common challenges and opportunities facing these groups but will also point the way towards novel solutions and courses of action.

The uniqueness of the symposium consists in its aim to provide recommendations on how to envision and improve the science-society interface.  As part of their involvement in the event, all speakers and participants will be asked to address the following question:

How can we understand and improve the interplay between science and society, and improve science policies for the future?

On the basis of the debate and answers, a results document will be created in which the potentially diverging views of different groups will be analyzed and distributed among media and key decision makers.

Science and Society 2013 aims to connect different communities and uncover common goals, competing concerns and the possibility of joint strategies. It will involve and reach out to practitioners from various sectors, academics of diverse disciplines and an increasingly interested public.  At its broadest level it will explore the relationships between public policy, scientific research and the study of science itself – including but limited to how these inform one another.

The symposium will have an academic component during much of the day; and a public component designed for a truly broad audience and potentially involving additional collaborators.

How?

The proposed Session Themes include:
Science and Democracy; Value-Laden Science; International Lessons in Science Policy; Citizen Science; Technology and Media; Responsible Innovation and the Future of Technology; Art, Science and Technology; Open Science; Government Science; Education and the Culture of Science; and Innovation and Society.

The event will produce the following outcomes:

  • New media and political interest, in particular with respect to key issues (e.g. muzzling scientists, evidence-based decision making, the importance of public science);
  • A results document, published by the ISSP, summarizing key insights regarding science and society for distribution among media and key decision makers;
  • New thinking and debate among scholars, policymakers, scientists, students and the public;
  • New networks;
  • Dissemination of conference content in print and/or www formats and/or video/podcast/live streaming;
  • Student training and engagement.

Why?

Science and technology shape our world. They present great promise but they are also the source of much controversy and social anxiety. Like never before, there is a need for broad and informed discussion of science and technology and their place in our society.

Yet the communities that engage in, benefit from, and seek to understand science and technology are often disconnected.  Their shared interests are often misunderstood, and their common goals overlooked.  This disconnect not only impoverishes our grasp of science and technology and their social implications but can also have negative consequences for the public good, particularly at a time when Canadian science faces such profound challenges.

Who?

The partners and co-organizers of the event are the Situating Science SSHRC Strategic Knowledge Cluster and the University of Ottawa Institute for Science, Society and Policy.

Situating Science, through the various conferences, symposium and public events it has supported across Canada with its many partners from different disciplines and sectors, has explored the social and cultural significance of science and technology.  And the ISSP has held and supported several events in Ottawa dealing with cutting-edge technologies and their social and political implications.

Both partners have brought diverse groups together before.  Each has its own networks, resources and strengths that align with select themes and audiences of the symposium.  The successful combination of these capacities will make Science and Society 2013 a multi-sectorial, multi-disciplinary event that addresses issues of concern to all Canadians.

War and Peace in an Age of Ecological Conflict

Dr. Bruno Latour is professor at Sciences Po Paris. Trained in philosophy, he has been instrumental in the development of an anthropology of science and technology. This field has had a direct impact on the philosophy of ecology and on an alternative definition of modernity. He has taught for many years in North American universities. Most of his books have been published with Harvard University Press. The most recently published is An Inquiry into Modes of Existence ‐ An Anthropology of the Moderns. While politics has always been linked to geography, the Earth itself has largely been seen as playing a backstage role, the mere window-dressing for human intention and interest. With the advent of the epoch known as the ‘Anthropocene’, the Earth is no longer in the background, but very much in the foreground, in constant rivalry with human intentionality. In the meantime, human action has taken on a dimension that matches that of nature itself, and consequently the definition of geo‐politics has been transformed. Appeals to nature, therefore, do not seem to have the same pacifying and unifying effect that they did in earlier ecological movements. By drawing on anthropological and philosophical literature, this lecture will discuss this new geopolitical framework and show how the extension of politics into nature must modify our views on war and peace in the future.
